We may build up to a total of 3 defense stations.

Does this mean we can build three additional defense stations to go along with the one we are automatically given at the start of round?

If one gets destroyed during the round, are we able to construct a replacement station?

Nope, three total for the round.
Once they blow, they are gone.

It looks like you can build one, but the “slot” is taken.
